Barnsley's reward for knocking holder Chelsea and Liverpool out of the FA Cup is a semifinal against fellow Championship (second division) side Cardiff City.
Monday's draw also pitted Portsmouth, the only Premier League side left in the competition, against Championship high-flyer West Bromwich Albion.
It is the first time since 1908 that only one team from the top flight reached the semifinals.
Both ties will be played at Wembley on the weekend of April 5-6.
In the Premier League, Arsenal was held to a 0-0 draw at Wigan. The Gunners have 66 points, leading Manchester United by two points, but the Red Devils have played one game less.
Everton kept up with Liverpool in the race for fourth place with a 1-0 win at Sunderland, and Tottenham beat 10-man West Ham 4-0.

FC Barcelona suffered a blow to its hopes of winning the Spanish league with a 2-1 defeat against visiting Villarreal on Sunday.
Marcos Senna converted a 31st-minute penalty and Jon Dahl Tomasson scored in the 81st for the third-place team, keeping Real Madrid eight points clear at the top. Real Madrid leads with 62 points, while Barcelona has 54, four more than Villarreal. Atletico Madrid and Santander each have 44.
